Short Description:

Antiphospholipid syndrome (APS) is a syndrome characterized by recurrent vascular embolism, miscarriage or intrauterine fetal death, thrombocytopenia, and neuropsychiatric symptoms, accompanied by persistent positivity for lupus anticoagulant and/or medium to high titers of anticardiolipin antibodies (ACA). Since medium to high titers of ACA are present in APS patients and persist over time, ACA serves as an important basis for diagnosing APS.

ACA is a group of heterogeneous antibodies with IgA, IgG, or IgM subtypes, among which IgG and IgM are the most common. Marai et al. tested 66 patients with primary APS and 60 patients with secondary APS, and found that the positive rate of ACA IgG was 25% to 33.3% and that of IgM was 1.6% to 7.5%; IgA antibodies were not detected separately. The positive rate of IgG-type ACA is the highest in patients with cerebral thrombosis, and it is closely associated with clinical manifestations. IgM-type ACA can serve as a prospective indicator for spontaneous abortion or stillbirth. Thrombocytopenia is more common in patients with IgG-type ACA and correlates with the severity of thrombocytopenia. Hemolytic anemia and neutropenia are associated with the IgM type.

Principle

This kit is designed to detect anti-cardiolipin antibodies in human serum or plasma samples. Polystyrene microwell strips are pre-coated with cardiolipin antigen. First, when serum or plasma specimens to be tested are added, the corresponding specific antibodies (cardiolipin-IgG-Ab and some IgM-Ab) present in the specimens bind to the antigens on the solid phase; other unbound components are then removed by washing. In the second step, horseradish peroxidase (HRP)-conjugated anti-human IgG specifically reacts only with anti-cardiolipin antibodies. After washing to remove unbound HRP conjugate, chromogen solutions are added to the wells. In the presence of the (cardiolipin Ag)-(cardiolipin-IgG)-(anti-human IgG-HRP) immune complex, TMB substrate is added for color development following plate washing. The HRP linked to the complex catalyzes the chromogen reaction, generating a blue substance. Upon adding 50μl of stop solution, the color turns yellow. The presence and concentration of cardiolipin-IgG antibodies in the sample are determined by measuring absorbance with a microplate reader.
