Anti-cyclic citrullinated peptide antibodies are autoantibodies that target cyclic citrullinated peptides, a type of modified protein antigen. Principle

This kit detects anti-cyclic citrullinated peptide antibodies (CCP antibodies) in human serum samples based on the indirect method, with purified cyclic citrullinated peptide antigens used as the coating antigen.

 

The testing process starts with adding the serum sample to reaction wells that have been pre-coated with the aforementioned purified antigens, followed by an incubation period. During this incubation, if CCP antibodies are present in the sample, they will specifically recognize and bind to the cyclic citrullinated peptide antigens coated on the microwells, forming stable antigen-antibody complexes. To ensure the accuracy of subsequent steps, unbound components in the reaction wells are removed through a washing process, which helps eliminate potential interference from other substances in the serum.

 

Next, enzyme conjugates are added to the reaction wells. After a second incubation, these enzyme conjugates will specifically attach to the existing antigen-antibody complexes, forming a larger immune complex that includes the antigen, antibody, and enzyme conjugate. When TMB substrate solution is introduced into the system, the enzyme in the conjugate catalyzes a chemical reaction with the TMB substrate, resulting in a visible color change. The intensity of this color reaction is directly related to the amount of CCP antibodies present in the original serum sample. Finally, a microplate reader is used to measure the absorbance (A value) of the reaction mixture. By analyzing this absorbance value, the level of CCP antibodies in the sample can be accurately determined, providing a reliable basis for relevant clinical testing and diagnosis.
