This kit is intended for the qualitative in vitro detection of anti-human chorionic gonadotropin antibodies (HCG-Ab) in human serum.

 

HCG-Ab is an autoantibody and ranks among the major causes of immunological infertility. Human chorionic gonadotropin (HCG), a pregnancy-specific hormone secreted by syncytiotrophoblasts, primarily functions to promote the development of the corpus luteum of pregnancy and the secretion of steroid hormones. It plays a crucial role in maintaining early pregnancy and counteracting the mother’s rejection of the fetus, serving as the key hormone for sustaining early fetal gestation.

 

HCG-Ab is mainly produced secondarily after miscarriages or HCG injections. Approximately 40% of individuals with a history of miscarriage test positive for HCG-Ab. When HCG-Ab binds to HCG, it blocks the active site of HCG and inhibits its physiological functions, making pregnancy unsustainable and easily leading to habitual or recurrent miscarriages, which in turn result in infertility. Its infertility-causing effect is supported by evidence such as difficulties in re-pregnancy after HCG injections and the contraceptive effect of HCG immunization.

Principle

This kit detects anti-human chorionic gonadotropin antibodies in human serum samples based on the indirect method, with purified human chorionic gonadotropin antigens used for pre-coating the microwells.

 

The testing process starts with adding the serum sample to the antigen-precoated reaction wells for incubation. If anti-human chorionic gonadotropin antibodies are present in the sample, they will specifically bind to the pre-coated antigens in the microwells, forming stable antigen-antibody complexes.

 

Next, enzyme conjugates are added. After a second incubation, these enzyme conjugates bind to the existing antigen-antibody complexes. When TMB substrate is introduced, a color reaction occurs under the catalysis of the enzyme. Finally, a microplate reader measures the absorbance (A value), which is used to determine the presence of anti-human chorionic gonadotropin antibodies in the sample.
