Anti-sperm (AS) Antibody ELISA Kit
Principle
This kit is designed to detect anti-sperm antibodies (ASAb) in human serum or plasma samples. Its polystyrene microwell strips are pre-coated with sperm antigens, forming the solid-phase reaction platform.
First, the serum or plasma specimens to be tested are added to the wells. If specific antibodies (such as AS-IgG-Ab and some AS-IgM-Ab) are present in the specimens, they will bind to the pre-coated sperm antigens on the solid phase. Unbound components are then removed through washing to eliminate interference.
In the second step, HRP (horseradish peroxidase)-conjugated anti-human IgG is added, which specifically binds to the anti-sperm antibodies already attached to the solid phase. After washing to remove unbound HRP conjugates, TMB substrate solution is added. The HRP in the formed immune complex (sperm antigen-anti-sperm antibody-HRP conjugate) catalyzes the TMB to develop a blue color. Adding 50μl of stop solution turns the color to yellow. Finally, a microplate reader measures the absorbance to determine the presence of AS-IgG antibodies in the sample.
