This product is designed for the qualitative in vitro detection of zona pellucida (ZP) antibody levels in human serum. The zona pellucida, a special extracellular matrix surrounding the oocyte, plays a vital role in sperm recognition, binding, and fertilization, as well as early embryonic development.

ZP antibodies are autoantibodies that target zona pellucida antigens. When they appear in the body, they can specifically bind to the zona pellucida, blocking the normal interaction between sperm and oocytes, thereby hindering fertilization. In addition, they may interfere with the implantation process of fertilized eggs, which is one of the important causes of autoimmune infertility.

Clinically, this detection is applicable as an auxiliary diagnostic method for autoimmune infertility. By detecting the level of ZP antibodies in patients’ serum, it can provide valuable reference information for clarifying the causes of infertility in some patients with unknown causes, helping doctors formulate more targeted diagnostic and treatment plans.

Principle

This kit detects zona pellucida antibodies (ZP-Ab) in human serum samples based on the indirect method, with purified zona pellucida used as the coating antigen.

The testing procedure begins by adding the serum sample to reaction wells pre-coated with the antigen, followed by incubation. If ZP-Ab is present in the sample, it will specifically bind to the coated zona pellucida antigen in the wells, forming stable antigen-antibody complexes.

Next, enzyme conjugates are added to the wells. After a second incubation step, these enzyme conjugates bind to the existing antigen-antibody complexes. When TMB substrate solution is introduced, a color reaction occurs under the catalytic action of the enzyme in the complex. Finally, a microplate reader is used to measure the absorbance (A value), which allows for the determination of ZP-Ab levels in the sample.
