Hepatitis A virus IgM (HAV-IgM) ELISA Kit
Principle:
This kit detects hepatitis A virus IgM antibodies (HAV-IgM) in human serum or plasma samples. The polystyrene microwell strips are pre-coated with antibodies directed against human immunoglobulin M proteins (anti-μ chain). Upon addition of the serum or plasma specimen, any IgM antibodies present in the sample are captured by the coated antibodies, while unbound components are removed by washing. In the second step, horseradish peroxidase (HRP)-conjugated HAV antigens are added, which specifically bind only to the captured HAV-specific IgM antibodies. After washing to remove unbound HRP-conjugates, a chromogenic substrate (TMB) is added to the wells. In the presence of the (anti-μ) - (HAV-IgM) - (HAV Ag-HRP) immunocomplex, the HRP enzyme catalyzes the substrate reaction, producing a blue color. Addition of 50 μL of stop solution halts the reaction and changes the color to yellow. The presence and concentration of HAV-IgM antibodies in the sample are determined by measuring the absorbance using a microplate reader.
