Short Description:

Enterovirus 71 IgM (EV71-IgM) ELISA kit is an enzyme-linked immunosorbent assay for the qualitative detection of IgM-class antibodies to Enterovirus 71 in human serum or plasma. It is intended to be used in clinical laboratories for diagnosis and management of patients related to infection with Enterovirus 71.

The human enterovirus 71 (EV71), the newest member of Enteroviridae, is a common cause for hand, foot and mouth disease (HFMD) and is sometimes associated with severe central nervous system diseases. Infections usually peak in June and July. An infection with EV71 can be both asymptomatic or may cause diarrhoea and rashes. Children under five are at higher risk to develop a serious infection than adults and older children.

EV71 is transmitted from person to person by direct contact and through contamination of hands or objects with infected faeces. Nose and throat secretions, saliva or fluid from blisters can also spread the virus.

EV71 can be isolated and detected in throat and stool specimens using PCR testing. Viral RNA has been detected in skin vesicle fluid, blood and urine. Earlier and easier diagnosis can be achieved using EV71-specific serological assays, including tests specific for IgM antibody.

Principle

This kit detects Enterovirus 71 IgM antibody (EV71-IgM) in human serum or plasma samples, polystyrene microwell strips are pre-coated with antibodies directed to human immunoglobulin M proteins (anti-μ chain).After first adding serum or plasma specimens to be examined, the IgM antibodies in the specimen can be captured, and other unbound components (including specific IgG antibodies) will be removed by washing. In the second step, the HRP (horseradish peroxidase)-conjugated antigens will specifically react only with EV71 IgM antibodies. After washing to remove the unbound HRP-conjugate, chromogen solutions are added into the wells. In presence of (anti-μ) - (EV71-IgM) - (EV71-Ag-HRP) immunocomplex, after washing the plate, the TMB substrate was added for color development, and the HRP connected to the complex catalyzes the color developer reaction to generate the blue substance, add 50μL of Stop Solution, and turn yellow. The presence of the absorbance of the EV71-IgM antibody in the sample was determined by a microplate reader.
