Short Description:

Hepatitis E virus IgM Test Cassette is used for qualitatively detection of Hepatitis E Virus IgM antibodies in human serum, plasma (EDTA, heparin, sodium citrate) or whole blood (EDTA, heparin, sodium citrate). The test is to be used as an aid in the diagnosis of viral hepatitis, which is caused by Hepatitis E Virus.

The Hepatitis E virus (HEV) is a non-enveloped, single stranded RNA virus primarily transmitted through the fecal-oral route, blood transfusion and possibly maternal-fetal. Infection with HEV causes acute sporadic and epidemic viral Hepatitis and induces acute or subclinical liver diseases similar to Hepatitis A. While there are four major genotypes of HEV, there is only one serotype.

HEV infection in humans produces IgM, IgA and IgG antibodies. HEV-IgM and HEV- IgA positivity is a sign of acute or recent HEV infection. Whether anti-HEV-IgM and anti-HEV-IgA are positive for one or both, they are indicative of recent HEV infection. The presence of recent HEV infection, combined with liver function, can be used to determine whether the infection is acute or recent. The presence of HEV infection in the liver can be used to determine whether the disease is acute hepatitis E or recovery from acute hepatitis E.

Principle

This kit detects hepatitis E virus IgM antibody (HEV-IgM) in human serum or plasma samples, polystyrene microwell strips are pre-coated with antibodies directed to human immunoglobulin M proteins (anti-μ chain). After first adding serum or plasma specimens to be examined, the IgM antibodies in the specimen can be captured, and other unbound components (including specific IgG antibodies) will be removed by washing. In the second step, the HRP (horseradish peroxidase)-conjugated antigens will specifically react only with HEV IgM antibodies. After washing to remove the unbound HRP-conjugate, chromogen solutions are added into the wells. In presence of (anti-μ) - (HEV-IgM) - (HEV Ag-HRP) immunocomplex, after washing the plate, the TMB substrate was added for color development, and the HRP connected to the complex catalyzes the color developer reaction to generate the blue substance, add 50μl of Stop Solution, and turn yellow. The presence of the absorbance of the HEV-IgM antibody in the sample was determined by a microplate reader.
